Mechanism of action

Plecanatide and its active metabolites bind to GC-C enzyme and act locally on the luminal surface of intestinal epithelial cells. The activation of GC-C enzyme increases the concentration of cyclic guanosine monophosphate (cGMP) both inside and outside the cell, while the increase in intracellular cGMP concentration is mainly achieved through the activation of ion channels of cystic fibrosis transmembrane regulator (CFTR), which stimulates the secretion of chloride and bicarbonate into the intestinal lumen, increasing and accelerating the passage of small intestinal fluid.
